Plant development is co mplex process associated with mult istage regulation. Chro mat in remodeling complexes which use energy for sliding or completely delet ing nucleosomes from the DNA chain play an essential ro le in development. Arabidopsis thaliana mutant bractea (bra) which is characterized by mult iple alterat ions in development such as dwarfis m and presence o f bracts and terminal flower was obtained and studied in our laboratory. Precise mapping using who le geno me sequencing showed that bra phenotype is caused by the nonsense mutation in CHB3 gene which encodes one o f DNAbinding subunits of chromat in remodeling co mplex SWI/SNF. The specificit y of DNA binding by SWI/SNF chromat in remodeling co mplex was analyzed. We performed a co mparison o f gene expressio n levels in wild t ype A. thaliana and in bra mutant using RNA-seq. The next step was to plot the densit y of differentially expressed genes along the chromoso mes. Densit y analysis revealed six regions 290 kb length where fraction o f different ially expressed genes was significant ly increased. Our findings evidence that CHB3 protein takes part in specific it y o f DNA binding by SWI/SNF chro mat in remodeling co mplex. Our future research will be aimed at the investigat ion o f other genes o f SWI/SNF co mplex in order to verify if there are other subunits which have specificit y to DNA binding.
